Glioblastoma multiforme (GBM) is an aggressive tumor characterized by high resistance to genotoxic therapies, frequently evading DNA damage through repair and tolerance pathways. Among these, the translesion synthesis (TLS) pathway, mediated by Y-family DNA polymerases, plays a fundamental role in the replication of genetic material even in the presence of lesions. Evidence suggests an intersection between genomic instability and proteotoxic stress through the Unfolded Protein Response (UPR). The present study aimed to investigate the impact of pharmacological POLH inhibition on the regulation of the IRE1/XBP1 axis of the UPR in a glioblastoma model. A172 cells constitutively expressing the fluorescent reporters XBP1-mNeonGreen and Apple-53BP1trunc were exposed to treatments with PNR-7-02, Olaparib, Thapsigargin, and Temozolomide. Reporter activation was monitored by time-lapse and end-point microscopy. Gene and protein expressions were quantified by RT-PCR and Western Blot, respectively. As a result, POLH inhibition generated a massive and rapid accumulation of the processed isoform of the XBP1 reporter, surpassing the level induced by the positive control thapsigargin. This hyperactivation occurred decoupled from the canonical UPR pathway, without an increase in the levels of the BiP chaperone or induction of the classical transcriptional target ERdj4. The expressive XBP1 splicing occurred in the absence of significant 53BP1 foci formation, demonstrating independence from massive double-strand breaks. Combination assays revealed that pre-sensitizing cells with PNR-7-02 synergistically potentiated the effect of TMZ. As a conclusion, POLH inhibition triggers a massive and non-canonical activation of XBP1 splicing, independent of endoplasmic reticulum stress activation. The data suggest a role for XBP1 as an integrator of cellular stresses, with potential regulation mediated by POLH. ...
