dc.description.abstractWetlands são sistemas que apresentam custo operacional e de implantação muito reduzidos se comparados à maioria dos sistemas de tratamento de efluentes. As comunidades microbianas presentes nesse sistema promovem a remoção de poluentes, da matéria orgânica e de nutrientes eutrofizantes. Este trabalho teve como objetivo a identificação das comunidades microbianas cultiváveis presentes no efluente e sua alternância ao longo do sistema em comparação aos dados físico-químicos. Amostras de efluente bruto e tratado foram coletadas no verão e no início do inverno de 2015 no sistema de wetlands construídos em um hotel. As amostras foram diluídas em série até a diluição 10-6 e as últimas três foram inoculadas em placas de Petri, em triplicata, contendo quatro meios de cultura. As amostras foram incubadas a temperatura ambiente (20°C ± 2°C) e 28ºC por 24-48 h. Foram obtidos 76 isolados bacterianos, sendo 54 das amostras de verão e 22 de inverno. Do total de isolados bacterianos obteve-se 18 Gram-positivas e 58 Gram-negativas. A média dos valores de nitrogênio total, nitrogênio amoniacal e fósforo demonstrou eficiência moderada de remoção pelo sistema de tratamento. Observou-se aumento de nitrato no efluente tratado nas duas coletas e um percentual de remoção maior no período de inverno para nitrogênio amoniacal e nitrogênio total. Dentre os 19 gêneros bacterianos identificados observou-se prevalência dos gêneros: Acinetobacter, Bacillus, Enterobacter, Klebsiella, Moraxella e Pseudomonas. As amostras representativas dos principais gêneros identificados bioquimicamente foram selecionadas para sequenciamento de DNA, cujo resultado confirmou os gêneros dos grupos microbianos.pt_BR
dc.description.abstractWetlands are systems that have low implementation and operational costs, when compared to other types of wastewater treatment systems. Microbial communities of wetlands systems promote the removal of pollutants, organic matter and eutrophication nutrients. This study aimed to identify the initial culturable microbial communities of the wetland effluent, as well as their changes throughout the system, and to compare this data with the physicochemical data. Raw and treated effluent samples were collected in the summer and early winter of 2015 in the wetlands system built in the hotel. Samples were serially diluted, up to 10-6 dilution and the last three dilutions were seeded on Petri dishes, in triplicate, into four culture media. After incubation at ambient temperature (20°C ± 2°C) and 28°C for 24-48 h a total of 76 bacterial isolates were obtained, 54 from summer and 22 from winter samples, of which 18 Gram-positive and 58 Gram-negative. The mean values of total nitrogen, ammoniacal nitrogen and phosphorus in the final treated effluent, showed a moderate efficiency of the wetlands system. Nitrate increased in the treated effluent in two samples, although a higher percentage of removal in the winter period for ammoniacal nitrogen and total nitrogen was registered. Among the 19 identified bacterial genera there was a prevalence of Acinetobacter, Bacillus, Enterobacter, Klebsiella, Pseudomonas and Moraxella. Samples that represented the majority of the genera identified biochemically, after being submitted for DNA sequencing confirmed the genera of the bacterial isolates.en
