dc.description.abstractA definição para abortamento é o término da gravidez antes de 24 semanas de gestação ou com peso fetal inferior a 500g. A incidência exata dos abortamentos não é conhecida devido à dificuldade em reconhecer concepções e perdas no início da gravidez. Abortamento Recorrente (AR) é definido por dois ou mais abortamentos consecutivos, sendo considerada uma enfermidade e não estando incluída entre os diferentes tipos de infertilidade. De todas as gestações reconhecidas, 25% resultam em abortamento sendo que, 5% dessas mulheres terão a experiência de ter dois abortamentos consecutivos e 1% terão três ou mais perdas. Em aproximadamente 50% dos casos de AR não é encontrada uma causa explicável, sendo então classificada como idiopática. Trombofilias consistem em desordens hemostáticas as quais levam a uma maior tendência a processos tromboembólicos, podendo também estar relacionadas a AR. O objetivo geral do presente trabalho é investigar mutações em genes relacionados a fenômenos de trombofilia em mulheres com duas ou mais perdas gestacionais. Nosso número amostral foi de 145 mulheres com no mínimo dois abortamentos consecutivos e 135 sem histórico de perdas gestacionais, oriundas do Hospital de Clínicas de Porto Alegre e do Hospital Fêmina. O DNA genômico foi obtido de amostra de sangue e de saliva. Os genótipos foram determinados através de real time PCR. As frequências encontradas dos alelos investigados no grupo de casos foram: 29,3% para o polimorfismo 677T do gene MTHFR (metilenotetrahidrofolato redutase), 1,0% para FVL (Fator V de Leiden) do gene FV, 0,4% para 20210A do gene FII (protrombina), 31,4% para -786C e 23,8% para 894T do gene eNOS (óxido nítrico sintase endotelial), as quais estão relacionados a trombofilias e têm sido associados a AR. Não encontramos diferenças estatisticamente significativas nas frequências genotípicas e alélicas dos polimorfismos estudados entre mulheres com AR e grupo controle. Também o hábito de fumar combinado à presença dos alelos de risco para as variantes aqui estudadas não parece ser determinante na enfermidade, mas o número de mulheres que relataram consumir álcool sugere que a substância pode estar relacionada à AR. Nossos dados não concordam com a proposição de que esses polimorfismos estejam relacionados com AR em nossa população.pt_BR
dc.description.abstractThe definition for abortion is the termination of pregnancy before 24 weeks of pregnancy or fetal weight less than 500g. The exact incidence of miscarriages is not known due to the difficulty in recognizing conceptions and losses at the beginning of the pregnancy. Recurrent Miscarriage (RM) is defined by two or more consecutive miscarriages, being considered an illness and is not included among the different types of infertility. Of all recognized pregnancies, 25% result in abortions and 5% of these women will have the experience of having two consecutive miscarriages and 1% has three or more losses. In approximately 50% of cases of RM is not found a cause that can be explained, and is then classified as idiopathic. Thrombophilias consist of haemostatic disorders which lead to a greater tendency to thromboembolic processes, and may also be related to the RM. The general objective of this study is to investigate mutations in genes related to the phenomena of thrombophilia in women with two or more miscarriages. Our sample size was 145 women with at least two consecutive miscarriages and 135 without a history of miscarriages, from the Hospital de Clínicas de Porto Alegre and the Hospital Fêmina. Genomic DNA was obtained from a sample of blood and saliva. The genotypes were determined by real time PCR. The observed frequencies of the polymorphisms investigated were: 29,3% to 677T MTHFR gene (methylenetetrahydrofolate reductase), 1,0% to FVL (Factor V Leiden) FV gene, 0,4% to 20210A FII gene (prothrombin), 31,4% to -786T and 23,8% to 894T eNOS gene (endothelial nitric oxide synthase), all of which are related to the thrombophilias and have been associated with RM. We found no statistically significant differences on genotypic and allelic frequencies studied polymorphisms between women with RM and control group. Also the habit of smoking combined with the presence of alleles of risk for the variants studied here does not seem to be the decisive factor in illness, but the number of women who reported consuming alcohol suggests that the substance is related to the RA. Our data do 7 not agree with the proposition that these polymorphisms are related with RM in our population.en
