dc.description.abstractA doença de Parkinson (DP) é uma desordem neurodegenerativa progressiva que atinge 1% da população mundial acima dos 55 anos. Em trabalho anterior, verificou-se que a atividade rotacional ipsilateral induzida pelo teste de motricidade sobre grade (TMG) é um parâmetro útil para a triagem de animais extensamente lesionados após infusão de 6-hidroxidopamina (6-OHDA) no feixe prosencefálico medial (FPM), um modelo animal muito utilizado para o estudo da doença. Entretanto, não se sabe como diferentes variáveis do aparato (altura e natureza da grade, presença ou não de paredes) e do modelo (diferentes sítios de infusão ou de variações no protocolo comportamental, com sessões de reteste ou aplicação de diazepam, interferem nessa atividade. Ratos Wistar machos de 110 dias de idade receberam infusões de 6-OHDA no corpo estriado (3,3 μg/μL, 6 μL), no FPM (3 μg/μL, 5,5 μL) ou na substância nigra (0, 3, 6 e 12μg/μL, 4 μL), sendo submetidos ao TMG 22 dias depois. No caso dos animais com infusão no FPM, o TMG teve variações de altura (8 ou 76,5 cm), de superfície (presença ou não de vidro transparente sobre a grade), ou de borda (presença ou não de parede a 30 ou 60 cm). Uma leva desses animais foi submetida a uma sessão prévia do TMG 7 dias antes da cirurgia, e outra recebeu diazepam (2 mg/kg, i.p.) ou apenas salina 30 minutos antes do teste. Verificou-se que a colocação de uma parede ao redor do aparato elimina a atividade rotacional (teste exato de Fisher, p=0, 016), embora não tenha sido possível observar esse efeito ao se discriminar a distância em 30 ou 60 cm (teste exato de Fisher, p= 0, 066 e p= 0,070, respectivamente). Não houve diferença significativa na atividade rotacional ao se variar a altura da grade ou sua superfície (teste exato de Fisher, p= 0,424 e p=1,000), nem com a administração prévia de diazepam (teste exato de Fisher, p= 0,648). Verificou-se que infusões realizadas no estriado podem produzir atividade rotacional no TMG (teste exato de Fisher, p=0,016) e que infusões na substância nigra o fazem quanto maior for a dose (teste de correlação de Spearman, ñ= 0,53, p=0,003). Os resultados sugerem que variações na altura da grade e na superfície de locomoção não interferem na indução da atividade rotacional no TMG, e que a mesma pode ocorrer em animais infundidos em quaisquer dos sítios de infusão utilizados neste trabalho. Entretanto, a possibilidade de tigmotaxia pela presença de paredes ao redor do aparato inibe essa atividade, embora a administração de diazepam não tenha surtido efeito, o que exige maiores estudos para se verificar o papel da ansiedade no processo. A exposição prévia à cirurgia, ao TMG não interferiu na atividade rotacional indicando que o fator novidade do teste parece não ser relevante para a atividade rotacional. Desse modo, destaca-se a tigmotaxia como principal fator interferente da atividade rotacional induzida pelo TMG e para o esclarecimento dos seus mecanismos e continuidade de estudos futuros utilizando o aparato.pt_BR
dc.description.abstractParkinson's disease (PD) is a progressive neurodegenerative disorder that affects 1% of the population above 55 years. In a previous work, it was shown that the footfault test is a tool for screening highly lesioned rats in modeling for PD using the toxin 6-hydroxydopamine (6-OHDA), which is widely used, by observing whether animals present or not context-induced ipsilateral rotational activity (CIIRA). However, it is not known whether CIIRA may change because of variations in the apparatus – such as in height or in surface texture, or in placing a surrounding wall - or of other differences, such as varying the 6-OHDA infusion site or the behavioral protocols, by including other testing sessions or applying a drug before testing. Male Wistar rats (110 days old) received 6-OHDA infusions into either the striatum (3.3 μg/μL, 6 μL), the MFB (3 μg/μL, 5,5 μL) or the substantia nigra (0, 3, 6, 12μg/μL, 4 μL), and were submitted to the footfault test 22 days later. In the case of animals that received infusions into the MFB, the footfault test varied in height (8 or 76.5 cm), surface (presence or not of a transparent glass on the grid), edge (presence or not of a surrounding wall 30 or 60 cm from the center). One cohort of these animals had a previous testing session 7 days before surgery and another received either diazepam (2 mg/kg, i.p.) or saline 30 min before testing. Our results show that a surrounding wall eliminates CIIRA (Fisher's exact test, p = 0.016), even though it was not possible to specify this effect concerning the wall distance from the center (30 or 60 cm; Fisher's exact test, p= 0.066 and 0.070, respectively). There was no difference by varying the height or the surface of the apparatus (Fisher's exact test, p =0.424 and 1.000, respectively) or by administrating diazepam before testing (Fisher's exact test, p =0, 648). It was observed that animals who received infusions into either the striatum (Fisher's exact test, p = 0.016) or the substantia nigra (Spearman correlation, ñ= 0,53, p=0,003) presented CIIRA in the footfault test. Our results suggest that CIIRA in the footfault is a robust effect in PD animal models considering the 6-OHDA infusion site and height of TMG, or if a transparent glass is placed on the grid. The presence of a surrounding wall eliminates the rotational activity, indicating that the possibility of presenting a tigmotaxic behavior interfere in this process. However, diazepam was not effective, indicating that the role of anxiety in this process requires further studies. Thus, we show the importance of tigmotaxic behavior as a main factor of rotational activity induced by the footfault test, and probably for understanding the underlying mechanisms and for the continuity of the studies using the apparatus.en
