dc.description.abstractO objetivo deste estudo foi investigar os efeitos do enriquecimento ambiental (EA) sobre a memória e a morfologia hipocampal, estriatal e cortical, estado oxidativo e níveis do fator neurotrófico derivado do encéfalo (BDNF), em ratos Wistar submetidos à hipóxia-isquemia (HI) neonatal. No capítulo 1 foram examinados os efeitos do EA sobre o prejuízo na memória observado no labirinto aquático de Morris (LAM) e sobre a atrofia hipocampal e cortical em animais hipóxicos-isquêmicos adultos. Ratos Wistar machos, no 7º dia pós-natal (DPN) foram submetidos ao modelo de Levine-Rice de HI neonatal, no qual a artéria carótida comum direita é ocluída permanentemente e, na seqüência, os animais foram mantidos em uma atmosfera hipóxica (90 min, 8% O2-92% N2). Duas semanas após a HI, os animais foram estimulados em um ambiente enriquecido (1h/dia, por 9 semanas). No LAM, os animais HI mantidos em ambiente padrão apresentaram pior performance quando comparados aos grupos CT e o grupo HI enriquecido teve mesmo comportamento que os grupos CT. Uma significante atrofia hipocampal e cortical foi identificada nos animais hipóxico-isquêmicos, porém, nesta análise, o EA não resultou em proteção significativa. O capítulo 2 teve como intuito investigar se a manutenção em ambiente enriquecido precocemente (8º-30 DPN) seria efetiva na recuperação da atrofia hipocampal pela HI, e, se os efeitos cognitivos vistos no EA diário seriam extensivos a outro tipo de memória e, também a outros períodos do desenvolvimento. Ratos e ratas Wistar machos foram submetidos à HI (conforme descrito acima) e, no dia seguinte, foram colocados em um ambiente enriquecido. Na seqüência, a memória de reconhecimento, no teste de reconhecimento de objetos e a memória espacial no LAM são analisadas. Na vida adulta, os animais foram submetidos a mais uma sessão experimental no LAM. O déficit cognitivo observado na memória de reconhecimento nos animais HI foi revertido pela estimulação. Porém, o prejuízo na memória espacial foi parcialmente revertido pelo EA; a memória de trabalho nas fêmeas, no período da adolescência, foi mais claramente beneficiada por este efeito. E, corroborando os resultados do primeiro experimento, a HI resultou em dano tecidual ao hipocampo e também ao estriado, sem indicação de efeito significativo pela estimulação. O capítulo 3 teve como objetivo investigar se a modulação do estado oxidativo e os níveis de BDNF cerebrais podem estar envolvidos no efeito neuroprotetor cognitivo do EA em animais adultos submetidos à HI. Ratos Wistar machos foram utilizados neste experimento; os procedimentos de HI e EA foram os mesmos do primeiro experimento. Os animais foram sacrificados 24 horas após o término do período de enriquecimento (85º DPN), quando hipocampo e córtex foram dissecados para a posterior determinação dos parâmetros de estresse oxidativo (quantificação de radicais livres, dano em macromoléculas e atividade da enzima superóxido dismutase - SOD) e dos níveis de BDNF. Os resultados demonstraram uma diminuição na quantidade de radicais livres no hipocampo esquerdo dos animais HI e um aumento destes níveis no córtex direito; e, também, um aumento na atividade da SOD no hipocampo direito dos animais HI e uma diminuição desta atividade no hipocampo esquerdo nos animais não estimulados. Além disso, os níveis de BDNF foram aumentados no hipocampo dos animais HI mantidos em ambiente padrão. Estes resultados sugerem que o efeito neuroprotetor cognitivo do EA diário pode não ser dependente da expressão de BDNF nem do estado oxidativo do hipocampo e córtex.pt_BR
dc.description.abstractThis study was undertaken to investigate the environmental enrichment (EE) effects over memory function and hippocampus and cortex morphology, oxidative status and brain-derived neurotrophic factor (BDNF) levels, consequent to neonatal hypoxiaischemia (HI) in Wistar rats. The objective of chapter 1 was to examine, in adulthood, the effects of daily EE on memory deficits in the Morris water maze (MWM) and hippocampal and cerebral damage, caused by neonatal HI. Male wistar rats at the 7th postnatal day were submitted to the Levine-Rice model of neonatal HI, comprising permanent occlusion of the right common carotid artery and a period of hypoxia (90 min, 8%O2-92%N2). Two weeks after the HI event, animals were stimulated by the enriched environment (1h/day for 9 weeks). In the MWM, non-stimulated HI animals had worse performance than controls and HI-enriched rats showed memory performance as good as that of controls. There was a reduction of both hippocampal volume and cortical area, ipsilateral to arterial occlusion, in HI animals; EE did not affect these morphological measurements, despite its clear cognitive neuroprotective effect. The chapter 2 investigated whether early continuous housing in an enriched environment would be effective in recovering hippocampal atrophy consequent to the neonatal HI, as well as if the benefits of continuous EE would extending to other memory types, both in adult and adolescent female and male rats. As described above, animals were submitted to HI and, starting 1day after the event, they were housed in an enriched environment (8th-30th PN day). Performances of animals in the novel-object recognition were assessed in the adolescence period and in the MWM task in the adolescence and, again, in adulthood. Cognitive deficits arising after neonatal HI in the novel-object recognition test were completely recovered by the environmental stimulation, in animals of both genders. However, spatial memory impairment in the MWM was partially prevented by EE; the effect was observed especially in female adolescent rats on the working memory. Corroborating the findings of the first experiment, there was no enrichment effect over hippocampus volume and striatum area. These data indicate that early housing in an enriched environment recovered performance in the object recognition task and, in adolescent females, in the working memory spatial task after a neonatal hypoxicischemic event. No effects of enrichment were revealed in adult animal performance nor in tissue atrophy of hippocampus and striatum consequent to HI. The chapter 3 was designed to investigate whether the modulation of brain oxidative status and/or BDNF content in adulthood are involved in functional neuroprotection caused by EE after neonatal HI. Protocols of the HI and EE utilized in this experiment were the same as of the first experiment. Male rats were sacrificed 24 hours after the enrichment period, i.e., at PN day 85. We have determined BDNF levels and several oxidative stress parameters, specifically the free radicals levels, macromolecules damage and superoxide dismutase (SOD) activity, in hippocampus and frontal cortex samples. It was found a decrease in free radical content in the left hippocampus of HI animals and an increase in non-stimulated rats in the right cortex; also, increased SOD activity in the right hippocampus of HI and a decrease of enzyme activity in the left hippocampus of stimulated groups were found. Moreover, BDNF levels were increased in the hippocampus of the non-stimulated HI group. These results suggest that the neuroprotective cognitive effect of daily environmental enrichment may not be dependent on BDNF expression nor on the oxidative status in hippocampus and cortex.en
