A acidúria 3-hidróxi-3-metilglutárica (HMGA), causada pela deficiência da enzima 3-hidróxi-3-metilglutaril-CoA liase (HL; EC 4.1.3.4), é um erro inato do metabolismo de herança autossômica recessiva. A HL catalisa a clivagem do 3-hidróxi-3-metilglutaril-CoA a acetoacetato e acetil-CoA, a qual é a última reação da síntese de corpos cetônicos e do catabolismo da leucina. Os indivíduos afetados pela HMGA apresentam acúmulo e elevada excreção urinária dos ácidos 3-hidróxi-3-metilglutárico (HMG), 3-metilglutárico (MGA), 3-metilglutacônico e 3-hidroxiisovalérico. Os pacientes apresentam predominantemente sintomas neurológicos, bem como hepatomegalia e cardiomiopatia, principalmente em situações de descompensação metabólica, quando ocorrem elevações drásticas nas concentrações dos metabólitos acumulados. Visto que os mecanismos fisiopatogênicos responsáveis pelo dano cerebral são pouco conhecidos, o objetivo do presente trabalho foi investigar os efeitos ex vivo da administração intraestriatal do HMG e do MGA sobre os parâmetros de estresse oxidativo substâncias reativas ao ácido tiobarbitúrico (TBA-RS), formação da carbonilas, concentrações de glutationa reduzida (GSH) e as atividades das enzimas antioxidantes glutationa peroxidase (GPx), glutationa redutase (GR), superóxido dismutase (SOD), catalase (CAT) e glicose-6-fosfato desidrogenase (G6PDH) em estriado de ratos jovens 30 min após a injeção. Nossos resultados demonstraram que a administração de HMG e MGA induziu peroxidação lipídica (aumento nos níveis de TBA-RS) e dano oxidativo proteico (formação de carbonilas) no estriado. Além disso, o HMG e o MGA diminuíram as concentrações de GSH, o mais importante antioxidante cerebral. Finalmente, as injeções de HMG e MGA aumentaram significativamente a atividade da GPx e diminuíram as atividades da SOD e da GR. Por outro lado, apenas o HMG aumentou a atividade da CAT e diminuiu a atividade da G6PDH. Nossos achados sugerem que o estresse oxidativo induzido in vivo pelo HMG e pelo MGA em estriado pode contribuir, pelo menos em parte, para a disfunção neurológica encontrada nos pacientes afetados pela HMGA. ...
