Abstract
The growing recognition of the interconnection between metabolic dysfunction and neuroinflammation reinforces the need to investigate, at the cellular level, the neuroprotective potential of gut microbiota-derived metabolites. The growing recognition of the interconnection between metabolic dysfunction and neuroinflammation reinforces the need to investigate, at the cellular level, the neuroprotective potential of gut microbiota-derived metabolites. This work aimed to investigate the cellular mechanisms and neuroprotective potential of Short-Chain Fatty Acids (SCFAs), acetate, propionate, and butyrate, in mitigating palmitate-induced lipotoxicity in in vitro cellular models of neurons (HT22 lineage) and microglia (BV2 lineage). Initially, a literature review was conducted on in vitro models used in the study of metabolic diseases associated with neurodegeneration, which validated the use of palmitate as a lipotoxic stress inducer. In the experimental stage, HT22 and BV2 cells were exposed to palmitate, either alone or co-incubated with SCFAs. The results demonstrated that palmitate induced cellular toxicity in the HT22 lineage, but not in BV2, at the tested concentrations. However, none of the SCFAs were able to significantly reverse the reduction in cell viability, the increase in late apoptosis, nor modulate lipid accumulation in the HT22 lineage. It is concluded that, although the lipotoxic insult model was validated, the therapeutic potential of SCFAs is highly dependent on factors such as the concentration used, the cell type, and the expression of specific receptors, which reinforces the need for optimization of in vitro models for the complete elucidation of their mechanisms of action. ...
