dc.description.abstractDesde 1982, o benzeno é classificado como agente químico carcinogênico e, portanto, sua utilização passou a ser controlada por órgãos regulamentadores mundiais, visando a redução dos níveis de exposição. Entretanto, o benzeno permanece sendo um importante poluente ambiental e estudos recentes mostram que seus efeitos tóxicos, especialmente hematotoxicidade, imunotoxicidade e carcinogenicidade, ocorrem mesmo em baixos níveis de exposição. Esse estudo buscou avançar no conhecimento relacionado ao mecanismo de toxicidade do benzeno, a fim de evidenciar potenciais biomarcadores precoces de dano. Bem como, avaliar a suscetibilidade individual através da avaliação de polimorfismos enzimáticos. Trabalhadores de postos de combustíveis do estado do Rio Grande do Sul, Brasil, compuseram o grupo exposto ocupacionalmente ao benzeno (GSW), e trabalhadores com atividades administrativas desse mesmo estado, compuseram o grupo não exposto ocupacionalmente (NEG). Avaliação da exposição ocupacional foi realizada através do monitoramento ambiental – quantificação dos níveis de benzeno no ar – e, quantificação do seu biomarcador de exposição ocupacional, o ácido trans, trans mucônico urinário (AttM). Foram observados maiores níveis de benzeno no ar e maiores níveis urinários de AttM nos indivíduos do grupo GSW comparados ao NEG. No capítulo I, avaliamos a imunotoxicidade e carcinogenicidade na exposição ocupacional ao benzeno, através da avaliação das principais moléculas coestimulatórias do sistema imune, B7.1 e B7.2, sistema complemento (SC) e gene supressor tumoral p53. Foi observada redução significativa na expressão proteica e gênica das moléculas B7.1 e B7.2 no grupo GSW comparado ao NEG. Além disso, os níveis das proteínas C3 e C4 do SC estavam reduzidas no grupo GSW, demonstrando prejuízo nessa outra via do sistema imune nos indivíduos expostos ao benzeno. Análise da expressão gênica do p53 demonstrou níveis reduzidos no grupo GSW, e modelo de correlação parcial demonstrou associações positivas entre as reduções das expressões gênicas das moléculas B7 e do p53. Esses resultados demonstram alteração em duas importantes vias de defesa do organismo frente ao desenvolvimento tumoral, e sugerem que a evasão imune juntamente com a supressão do p53 têm um papel importante na carcinogenicidade do benzeno. No capítulo II, avaliamos a influência de polimorfismos enzimáticos (GSTM1, GSTT1, CYP2E1 e ALAD) sobre parâmetros hematológicos, atividade enzimática da δ- 14 aminolevulinato desidratase (δ-ALA-D) e biomarcadores de estresse oxidativo. Observamos redução na atividade da δ-ALA-D no grupo GSW quando comparado ao NEG, corroborando com resultados previamente publicados por nosso grupo. A inibição da δ-ALA-D estava associada negativamente a maior tempo de exposição, maiores níveis urinários de AttM e de benzeno no ar, demonstrando a ação do benzeno sobre a inibição dessa enzima. A inibição da δ-ALA-D pode resultar em anemia, um efeito hematotóxico comum na exposição ao benzeno, portanto, sugere-se a avaliação da δ-ALA-D como potencial biomarcador precoce de hematotoxicidade do benzeno. De fato, a inibição da δ-ALA-D estava correlacionada positivamente a menores níveis de hemoglobina (Hb) e hematócrito (HCT) observados nos individuos do grupo GSW. Análise genotípica observou aumento significativo de carreadores do genótipo polimórfico do gene ALAD no grupo GSW comparado ao NEG, e apesar do aumento observado de genótipos polimórficos nos genes GSTT1 e CYP2E1 no grupo GSW, não foram observadas diferenças significativas entre os genótipos desses genes e do GSTM1 entre os grupos. Agrupamento genotípico revelou que indivíduos portadores de genótipo polimórfico do gene ALAD apresentaram menor atividade de δ-ALA-D, sugerindo, pela primeira vez, a ocorrência desse polimorfismo como potencial biomarcador de suscetibilidade aos efeitos hematotóxicos do benzeno. Além disso, a inibição dessa enzima resulta no acúmulo do seu substrato, um composto pró-oxidante, que pode contribuir para o aumento do estresse oxidativo, o qual foi observado nesse estudo através de marcadores clássicos como malondialdeído; níveis de grupamentos tiólicos não proteicos e enzimas do sistema antioxidante no grupo GSW comparado ao NEG. Portadores de genótipos polimórficos nos genes GSTT1 e CYP2E1 apresentaram aumento na atividade enzimática da glutationa peroxidade (GPx) e glutationa transferase (GST), respectivamente, demonstrando um possível efeito protetivo desses polimorfismos. Nossos resultados sugerem potenciais biomarcadores precoces de hematotoxicidade, imunotoxicidade e carcinogenicidade na exposição ocupacional ao benzeno, e adicionalmente, evidenciamos o polimorfismo do gene ALAD como um potencial biomarcador de suscetibilidade, entretanto estudos adicionais são necessários para confirmar esses achados prévios.pt_BR
dc.description.abstractBenzene was classified in 1982 as a carcinogenic chemical agent and its use has been controlled ever since by regulatory agencies worldwide, aiming to reduce exposure levels. However, benzene remains as an important environmental pollutant and recent studies show that its toxic effects, especially hematotoxicity, immunotoxicity and carcinogenicity, occur even at low levels of exposure. This study aimed to further the knowledge on the mechanism of benzene toxicity in order to evidence potential early biomarkers of damage, and to evaluate the individual susceptibility through the evaluation of enzymatic polymorphisms. Gas station workers from the Brazilian state of Rio Grande do Sul were recruited as the group of individuals occupationally exposed to benzene (GSW), while workers with administrative activities from the same region composed the non-occupationally exposed group (NEG). Occupational exposure was assessed by means of the environmental monitoring accomplished by quantification of indoor air benzene in the workplace; in addition, the urinary biomarker trans, trans, muconic acid (t, t - MA) was determined to measure the occupational exposure to benzene. There were higher airborne benzene levels and higher urinary levels of t, t - MA in individuals of the GSW group compared to NEG. In chapter I, we evaluated the immunotoxicity and carcinogenicity of benzene occupational exposure through the evaluation of the major costimulatory molecules of the immune system, B7.1 and B7.2, complement system (CS) and the tumor suppressor gene p53. Significant reduction in protein and gene expression of B7.1 and B7.2 molecules was observed in the GSW group compared to NEG. Besides, levels of C3 and C4 complement proteins were reduced in the GSW group, demonstrating that the impairment in this additional pathway of the immune system occurs in subjects exposed to benzene. Analysis of the p53 gene expression demonstrated reduced levels in the GSW group, and partial correlation model demonstrated positive associations between the gene expression reductions of the B7 molecules and p53. These results demonstrate alterations in two important pathways of organism defense against tumor development, and suggest that immune evasion together with p53 suppression have an important role in the carcinogenicity of benzene. In the chapter II, we evaluated the influence of enzyme polymorphisms (ALAD, CYP2E1, GSTM1 and GSTT1) on hematological parameters, 16 enzymatic activity of δ-aminolevulinate dehydratase (δ-ALA-D) and oxidative stress biomarkers. We observed a reduction in the δ-ALA-D activity in the GSW group when compared to the NEG, corroborating with results previously published by our group. Inhibition of δ-ALA-D was negatively associated with higher exposure time, higher levels of t, t - MA and airborne benzene, demonstrating the action of benzene on the inhibition of this enzyme. Inhibition of δ-ALA-D may result in anemia, a common hematotoxic effect of benzene exposure, therefore the evaluation of δ-ALA-D is suggested as a potential early biomarker of benzene hematotoxicity. In fact, inhibition of δ-ALA-D was positively correlated with lower levels of hemoglobin (Hb) and hematocrit (HCT) observed in individuals from GSW group. Genotypic analysis showed a significant increase in the ALAD gene polymorphism in the GSW group compared to NEG. Despite the observed increase of polymorphic GSTT1 and CYP2E1 genotypes in the GSW group, no significant differences were observed between the genotypes of these genes and GSTM1 between groups. Genotypic grouping revealed that individuals carrying polymorphic genotypes of the ALAD gene had lower δ-ALA-D activity, suggesting for the first time the occurrence of this polymorphism as a potential susceptibility biomarker to the hematotoxic effects of benzene. In addition, inhibition of this enzyme results in the accumulation of its pro-oxidant substrate, which may contribute to the increase of oxidative stress observed in this study by classic markers such as malondialdehyde, non-protein thiol groups and enzymes of the antioxidant system in the GSW group compared to NEG. Polymorphic genotype carriers of GSTT1 and CYP2E1 genes showed an increase in the enzymatic activity of glutathione peroxidase (GPx) and glutathione S-transferase (GST), respectively, demonstrating a possible protective effect of these polymorphisms. Our results suggest potential early biomarkers of benzene occupational exposure, and additionally evidence a potential biomarker of susceptibility, however additional studies focused on the results obtained are necessary to confirm these findings.en
