Caracterização genotípica de variações intraespecíficas e padrões de polimorfismo de Bipolaris sorokiniana utilizando RFLP-ITS

dc.description.abstract | O fitopatógeno Bipolaris sorokiniana infecta diversas gramíneas causando moléstias como a podridão comum da raiz, carvão do nó, mancha marrom e ponta preta dos grãos. Dentre as gramíneas infectadas encontramos a cevada, o trigo, triticale e outros. O trigo representa, no âmbito mundial, a segunda maior cultura de grãos, no entanto, o Brasil está entre os três maiores importadores mundiais de trigo. A principal fonte de inóculo do fitopatógeno é a semente, contudo a elevada variabilidade morfológica e fisiológica de B. sorokiniana dificulta a sua identificação, assim como o tratamento das sementes infectadas. Sendo assim, a caracterização molecular do fitopatógeno se mostra de fundamental importância. O objetivo desse trabalho foi estudar a diversidade genotípica de B. sorokiniana isolados de sementes de trigo do Brasil e de outros países utilizando PCR-RFLP das regiões ITS do rDNA ribossomal. Para tanto, quarenta isolados polispóricos foram utilizados. Foi realizada a extração, purificação e quantificação do DNA genômico e as reações de amplificação foram realizadas com os oligonucleotídeos iniciadores universais para fungos ITS4 e ITS5. Todos os isolados apresentaram um produto de amplificação com aproximadamente 600pb, esperado para as regiões ITS, junto com o segmento 5.8S do rDNA. Os produtos das amplificações foram digeridos com a endonuclease de restrição EcoR I e os produtos de amplificação e clivagem submetidos à eletroforese em gel não desnaturante de poliacrilamida 8% para posterior análise dos perfis dos fragmentos de DNA. Os dados obtidos a partir da RFLP das regiões ITS1, 5.8S e ITS2 dos isolados revelaram a presença de polimorfismo intra-específicos. | pt_BR |
dc.description.abstract | The pathogen Bipolaris sorokiniana infects several grasses causing diseases such as common root rot, charcoal node, brown spot and black point of the grains. Among the grasses found infected barley, wheat, triticale and others. The wheat represents, at the global, the second largest grain crop, however, Brazil is among the world's three largest importers of wheat. The main source of inoculum of the pathogen is the seed, but the high variability of morphological and physiological B. sorokiniana difficult to identify them, as well as the treatment of infected seeds. Thus, the molecular characterization of the pathogen be essential. The aim of this work is to study the genotypic diversity of B. sorokiniana isolated from wheat seeds from Brazil and other countries using RLFP-ITS. For this, forty isolates were used. It made the extraction, purification and quantification of genomic DNA and amplification reactions were performed with universal primers for fungi ITS4 and ITS5. All isolates showed an amplification product of approximately 600pb, expected for the ITS region, along with the 5.8S rDNA segment. The products of amplification were digested with restriction endonucleases EcoR I and Hind III and the amplification products were subjected to cleavage and gel electrophoresis nondenaturing 8% polyacrylamide for further analysis of the profiles of DNA fragments. The data obtained from the RFLP regions ITS1, 5.8S and ITS2 of the isolates revealed the presence of intra-specific polymorphism. | en |
