Os sistemas nanoestruturados estão sendo cada vez mais pesquisados e estudados devido à necessidade cada vez maior de aperfeiçoar a ação de fármacos no organismo. Estes sistemas trazem diversas vantagens, tais como controlar a liberação de fármacos e possivelmente diminuir a irritação de substâncias irritantes quando aplicadas sobre a pele. A capsaicina, um fármaco analgésico usado no tratamento da dor de origem neuropática provoca irritação quando aplicada na via tópica. O uso de formulação comercial desta substância produz irritação nos usuários, motivo pelo qual muitos abandonam o tratamento. Uma alternativa promissora para a diminuição deste efeito indesejável na pele consiste da nanoencapsulação desse fármaco o que poderia aumentar a adesão ao tratamento. Portanto, esse trabalho avalia o poder irritante em humanos dessa nova formulação tópica composta de hidrogel de quitosana contendo nanocapsulas de capsaicina quando comparada com a formulação comercial e com uma fornulação semelhante contendo a capsaicina na forma livre (n=19 voluntários). Foram levantados dados qualitativos sobre a intensidade da sensação produzida nos voluntários e dados quantitativos do nível de eritema, pH e taxa de hidratação. Os dados foram avaliados por meio de estatística descritiva e ANOVA para medidas repetidas. Os resultados encontrados demonstram que a formulação desenvolvida com nanocapsula diminui significativamente o efeito irritante nos voluntários quando comparada com a formulação comercial e também se comparada a formulação livre, sem levar a mudanças significativas da hidratação da pele e com diferenças significativas para o pH. Conclui-se também que o veículo inovador proposto composto de quitosana e nanocápsulas poliméricas não apresentou efeito significativo de irritação nos voluntários, apresentando-se como alternativa promissora para aplicação de capsaicina ou outros ativos na pele. ...
