A enzima glutamina sintetase (GS), peça-chave no metabolismo do nitrogênio, apresenta papel primordial na biossíntese de L-glutamina e assimilação desse elemento para a grande maioria das bactérias. GS atua como catalisadora da reação que forma L-glutamina a partir de ácido L-glutâmico e amônio, sendo necessário gasto de ATP para tal. Dessa maneira, o íon amônio é introduzido ao metabolismo celular pela GS e pela glutamato sintase, produzindo L-glutamina e L-glutamato, respectivamente. O gênero Paenibacillus é alvo de diversos estudos; no entanto, ainda se fazem necessárias mais análises genômicas, tendo em vista sua importância para a biotecnologia e o agronegócio, ampla distribuição, função nas comunidades microbianas e fixação de nitrogênio, observada em várias espécies do gênero. Paenibacillus riograndensis (SBR5), sinônimo heterotípico posterior de P. sonchi (X19-5 T), bactéria isolada de trigo (Triticum aestivum), quando comparada a outras do gênero apresenta peculiaridades. SBR5 contém, em seu genoma, três genes codificadores de proteínas homólogas à GS (nomeadas no presente estudo como “GSs-like” – GSL1, GSL2 e GSL3). Este estudo visa à caracterização das funções de tais proteínas. Para a expressão das três proteínas (GSL1, GSL2 e GSL3) foi utilizada Escherichia coli BL-21 e o plasmídeo pASKIBA3, a qual foi confirmada em gel de poliacrilamida (SDS-PAGE). Utilizando-se o resina (meio) StrepTactinTM SepharoseTM High Performance (GE Healthcare), as proteínas foram purificadas por cromatografia de afinidade. Após a purificação e diálise das proteínas, as mesmas foram quantificadas pelo método de Bradford e armazenadas à -20°C, para posterior análise. Foi utilizado o método colorimétrico baseado na liberação de fosfato inorgânico (Pi) resultante da hidrólise de ATP para medição da atividade biossintética das GSs-like. Observou-se atividade de 11,6 e 9,63 nmol Pi/mg para GSL1 e GSL2, respectivamente, enquanto GSL3 não apresentou atividade biossintética. Tendo em vista uma possível semelhança entre as GSs em questão e outras proteínas caracterizadas na literatura, foi realizado um ensaio colorimétrico para verificar atividade na presença de poliaminas como substrato, em substituição ao amônio. Foi observada atividade para as três GSs-like, em diferentes níveis, mostrando a capacidade dessas enzimas em utilizar poliaminas como fonte de nitrogênio. Testando-se possíveis inibidores foi notada inibição de GSL1 apenas por glutamina, sendo as outras proteínas estimuladas pelos demais substratos avaliados. ...

The enzyme glutamine synthetase (GS), a key part in nitrogen metabolism, plays a major role in L-glutamine biosynthesis and assimilation of this element for the vast majority of bacteria. GS acts as a catalyst for the reaction that forms L-glutamine from L-glutamic acid and ammonium, requiring ATP expense. Thus, the ammonium ion is introduced to cellular metabolism by GS and glutamate synthase, producing L-glutamine and L-glutamate, respectively. The genus Paenibacillus is the target of several studies; however, further genomic analysis is still needed, given its importance for biotechnology and agribusiness, broad distribution, microbial function and nitrogen fixation, observed in several species of the genus. Paenibacillus riograndensis (SBR5), posterior heterotypic synonym of P. sonchi (X19-5 T), bacterium isolated from wheat (Triticum aestivum), when compared to others of the genus presents peculiarities. SBR5 contains in its genome three genes coding for GS homologous proteins (named in the present study as "GSs like" - GSL1, GSL2, and GSL3). This study aims to characterize the functions of such proteins. For expression of the three proteins (GSL1, GSL2, and GSL3), Escherichia coli BL-21 and plasmid pASK-IBA3 were used, and their expression was confirmed on polyacrylamide gel (SDS-PAGE). Using StrepTactin™ Sepharose™ High Performance resin (medium) (GE Healthcare), proteins were purified by affinity chromatography. After protein purification and dialysis, they were quantified by the Bradford method and stored at -20°C for later analysis. The colorimetric method based on the release of inorganic phosphate resulting from ATP hydrolysis was used to measure the biosynthetic activity of GSs-like. Activity of 11.6 and 9.63 nmol Pi/mg was observed for GSL1 and GSL2, respectively, while GSL3 showed no biosynthetic activity. In view of a possible similarity between the GSs in question and other proteins characterized in the literature, a colorimetric assay was performed to verify activity in the presence of polyamines as substrate, replacing ammonium. Activity was observed for the three GSs-like, at different levels, showing the ability of these enzymes to use polyamines as nitrogen source. Testing for possible inhibitors, GSL1 inhibition was noted only by glutamine, and the other proteins were stimulated by the other substrates evaluated. ...
