RESUMO Atualmente, já está bem estabelecido que o principal fator de risco para o desenvolvimento do câncer de colo de útero é a infecção pelo papilomavírus humano (HPV). Entre os tipos de HPV mais frequentes, podemos citar o HPV 16 e o HPV 18, os quais são responsáveis por aproximadamente 70% dos casos de câncer de colo de útero. A proliferação descontrolada e acelerada de células anormais observada em neoplasias requer uma reprogramação metabólica para suprir as necessidades energéticas utilizadas na divisão celular. Proteínas, lipídios e ácidos nucleicos são fundamentais para que a célula tumoral possa proliferar. A síntese de novo de ácidos graxos surge como um mecanismo importante no fornecimento de lipídios. Este processo ocorre no citoplasma das células e é realizado pela enzima metabólica ácido graxo sintase (FASN) através de reações de condensação entre acetil CoA e malonil CoA. Ainda, a proteína FASN possui um papel importante na homeostase energética, pois converte o excesso de carbono ingerido em ácidos graxos que podem ser utilizados para produzir energia através da b oxidação. A FASN contribui para a produção de lipídios de membrana através da formação do seu principal produto, o palmitato. Devido a dieta rica em ácidos graxos, a síntese de novo e a atividade da FASN encontram se em níveis baixos na maioria dos tecidos, com exceção do fígado, cérebro, tecido adiposo, pulmão e glândulas mamárias lactantes. Estudos tem demonstrado que muitas neoplasias malignas apresentam níveis elevados de FASN e que sua inibição, específica ou farmacológica, resulta em atividade antitumoral. A fármaco orlistate (ORL), foi originalmente desenvolvida para o tratamento da obesidade pois é um inibidor de lipases pancreáticas no trato gastrointestinal. No entanto, este composto mostrou ser um inibidor irreversível de FASN ao se ligar ao domínio tioesterase (TE), o qual é responsável pela liberação do palmitato. Neste trabalho foram utilizadas diferentes linhagens celulares de câncer de colo de útero (C 33A, ME 180, HeLa e SiHa), com objetivo de estudar os efeitos da inibição da FASN sobre a proliferação celular e indução de apoptose. As linhagens celulares foram tratadas com diferentes concentrações de ORL e estudos de proliferação e determinação de morte celular foram realizados através de contagem celular e marcação com Anexina V e iodeto de propídio. Assim como para outras linhagens celulares de diferentes neoplasias malignas, a inibição da FASN pelo ORL foi capaz de diminuir a viabilidade celular e causar apoptose em todas as linhagens celulares. No entanto, as linhagens SiHa e HeLa, que apresentaram uma maior expressão de FASN, foram também as linhagens em que a inibição da proliferação foi maior quando comparada com as linhagens C 33A e ME 180. Entre todas as linhagens estudadas a que menos expressou FASN foi a linhagem ME 180, que também mostrou ter menor redução na viabilidade celular. Também foi possível verificar que a diminuição na viabilidade ocorreu por apoptose e não por necrose, visto que, a porcentagem de células em necrose foi baixa em todas as linhagens após o tratamento com ORL. O presente trabalho, demonstrou pela primeira vez que diferentes linhagens celulares de câncer de colo de útero expressam FASN de forma desigual e que podem ser afetadas pela inibição farmacológica desta com ORL. ...
