dc.description.abstractCryptococcus neoformans e Cryptococcus gattii são leveduras encapsuladas causadores da criptococose. O tratamento da doença apresenta elevada toxicidade, fazendo-se necessário o desenvolvimento de fármacos menos tóxicos e mais eficientes. Com esse propósito, nosso grupo de pesquisa identificou a presença de dois iridóides com ação antifúngica no extrato de sementes de Allamanda polyantha: a plumieridina e o plumierideo. Relatos anteriores demonstraram a conversão do plumierideo a plumieridina por hidrólise enzimática, entretanto, seu alvo molecular ainda não é conhecido. Dessa forma, esse trabalho tem como objetivo identificar, por métodos in silicio, possíveis proteínas alvo para a ação da plumieridina em C. neoformans e C. gattii. Para tanto, utilizamos o servidor do PharmMapper para predizer possíveis alvos protéicos desse ligante. Dentre esses, identificamos algumas enzimas da rota das pirimidinas, selecionando a timidilato sintase (TS), um dos alvos da ação do antifúngico flucitosina, para iniciar nossos estudos. As TSs das duas espécies foram modeladas, e o docking da molécula da plumieridina foi realizado nas estruturas resultantes. Alguns complexos TS-plumieridina foram selecionados e simulados por dinâmica molecular, assim como as TSs das duas espécies na ausência do ligante. A plumieridina, no entanto, permaneceu interagindo com a TS apenas nas simulações dos complexos onde ela se encontrava no sítio catalítico. Foi possível observar divergências entre as simulações das TSs das duas espécies com a plumieridina. A de C. neoformans demonstrou maiores modificações estruturais, tendo ocorrido o desenovelamento de estruturas secundárias porém, fora isto, a forma de interação da plumieridina com a TS apresentou pouca variação ao longo da simulação. Em contrapartida, em C. gattii foi observado grande conservação da estrutura secundária, assim como uma variação significante na interação TS-plumieridina, ocorrendo um deslocamento do ligante no sítio catalítico. Apesar disso, em ambos os casos a plumieridina interagiu com regiões essenciais na catálise enzimática, indicando potencial para bloquear o sítio catalítico e impedir a ligação dos substratos, inviabilizando, dessa forma, o correto funcionamento da enzima.pt_BR
dc.description.abstractCryptococcus neoformans and Cryptococcus gattii are encapsulated yeast, and the main cause of cryptococcosis. Due to their high toxicity, is necessary to develop more effective drugs to treat cryptococcosis. With this purpose, our group has identified the presence of two potential antifungal iridoids in seed extracts of Allamanda polyantha: the plumieridin and the plumieride. Previous reports demonstrate the conversion of plumieride to plumieridin, by enzymatic hydrolysis, however, its target is not yet known. Thus, this study aims to identify, by in silicio methods, possible target proteins for the action of plumieridin in C. neoformans and C. gattii. For this, we used PharmMapper server to predict possible targets of this ligand. Among the results, we identified some enzymes from the pyrimidines pathway, selecting thymidylate synthase (TS), one of the targets of flucytosine action, in order to initiate our work. The enzymes from both species were modeled, and the docking of plumieridin’s molecule was performed on the resulting structures. Some TS-plumieridin complexes were selected and simulated by molecular dynamics, as well as the TSs from the two species without ligand. The plumieridin, however, remained interacting with the TS only in the simulations where it was at the catalytic site. Divergences between the simulations of TSs from the two species with plumieridin were observed. C. neoformans demonstrated major structural changes, including the unfolding of some secondary structure. Furthermore, plumieridin interaction with TS showed little variation during the simulation. In contrast, for C. gattii there was an extensive conservation of secondary structure, as well as a significant variation in TS-plumieridin interaction with a displacement of the ligand in the catalytic site. Despite this, in both cases the plumieridin interacted with essential regions for enzyme catalysis, indicating a potential to block the catalytic site preventing binding of substrates and affect the correct function of the enzyme.en
