Introdução: A OMS estima que as infecções causadas por G. lamblia sejam a doença protozoária intestinal mais frequentemente relatada em todo o mundo. No entanto, um problema comum aos medicamentos usados no tratamento da giardíase diz respeito às linhagens resistentes. Sendo assim, buscam-se novos fármacos capazes de combaterem as infecções causadas por G. lamblia, através da utilização de enzimas validadas e específicas do parasita. A enzima guanina fosforibosil transferase (GPRTase) é essencial para a produção de nucleotídeos no parasita, uma vez que os parasitas, diferentes dos humanos, são incapazes da síntese de novo de nucleotídeos. Objetivos: Identificar novos inibidores da enzima GPRTase de Giardia lamblia que possuam eficácia e perfil farmacocinético adequado para serem usados no tratamento da giardíase. Material e métodos: Primeiramente, estudou-se o sítio de ligação das enzimas GPRTase de G. lamblia e de sua homóloga humana, a hipoxantina-guanina fosforibosil transferase (HGPRTase). Em seguida, construiu-se um modelo farmacofórico para se triar estruturas químicas no banco de dados da ZINC que possuam complementaridade com o sítio da GPRTase. Ademais, realizou-se o processo de docagem molecular dessas estruturas utilizando o complexo cristalográfico da GPRTase em complexo com o inibidor do estado de transição, da enzima (PDB ID: 1DQN), por meio da utilização dos programas Glide e Gold, abrangendo as etapas de validação, docagem e docagem consensual. Por fim, realizou-se a predição das propriedades farmacocinéticas dos melhores ligantes através do programa Volsurf+. Resultados e Discussão: O estudo do sitio de ligação da enzima GPRTase demonstrou que as interações com os resíduos Trp180, Leu181, Asp187 parecem ser importantes para a especificidade da enzima. Além disso, a possibilidade de interação com Lys152 poderia aumentar a seletividade frente à GPRTase. O modelo farmacofórico de quatro pontos resultou na obtenção de 1230 estruturas do banco de dados da ZINC. A docagem consensual utilizando Glide-Glidescore e Gold-GoldScore não apresentou consenso visual das poses. Foram escolhidas 5 estruturas avaliadas pelo GoldScore para predição farmacocinética, a qual demonstrou que os ligantes parecem não serem absorvidos pelo intestino e não ultrapassam a barreira hematoencefálica. Conclusões: Não foi possível obter um consenso visual das poses, devido ao tamanho dos ligantes e número de ligações rotacionáveis. Tendo em vista os resultados obtidos, novos procedimentos serão realizados visando o aumento do número de poses da docagem, docagem totalmente flexível e triagem virtual com nova base de dados através de novo modelo farmacofórico. ...
