O estudo da interação entre ligantes e seus receptores-alvo tende a iniciar pela caracterização da conformação adotada pelos determinados compostos. O estudo da interação entre ligantes e seus receptores-alvo tende a iniciar pela caracterização da conformação adotada pelos determinados compostos. Dessa forma, as saponinas, que são glicosídeos formados por uma porção hidrofílica, composta por carboidratos, e uma lipofílica, composta por um triterpeno ou um esteróide, foram identificadas como moléculas que possuem diversas ações e atividades biológicas. Entretanto, ainda existem muitas dificuldades na elucidação estrutural desses compostos, principalmente devido à alta flexibilidade encontrada nas regiões compostas por sacarídeos. Desafios adicionais ainda incluem o fato de muitas saponinas terem suas estruturas elucidadas em solventes não-aquosos, para o caso das saponinas Erucasaponina A e Estelatosídeo B, e a instabilidade química e a formação micelar, no caso da Quillaja saponaria – 21. Dessa forma, o presente trabalho emprega técnicas de modelagem molecular para realizar a caracterização conformacional em nível atômico de quatro saponinas (Erucasaponina A, Estelatosídeo B, Chikusetsusaponin IVa e Quillaja saponaria – 21), utilizando mapas de contorno de energia e simulações de dinâmica molecular para refinamento dos modelos. Os modelos tridimensionais das saponinas Erucasaponina A e Estelatosídeo B foram devidamente validados em solvente aromático (piridina) e um modelo adicional para cada uma foi proposto em ambiente fisiológico. A Chikusetsusaponin IVa e a Quillaja saponaria – 21 tiveram modelos construídos também, sendo que a formação micelar da última pode ser observada em solução aquosa e validada com dados experimentais. Assim, espera-se que os modelos obtidos demonstrem a força dos métodos computacionais na caracterização de biomoléculas e possam contribuir para futuras pesquisas envolvendo glicoconjugados. ...
