A Tacrina foi o primeiro composto aprovado como fármaco para o tratamento da Doença de Alzheimer (DA). Recentemente nosso grupo de pesquisa desenvolveu uma nova metodologia para a obtenção de análogos quirais da Tacrina a partir de cetonas terpênicas, utilizando ácidos de Lewis como catalisadores. A Tacrina foi o primeiro composto aprovado como fármaco para o tratamento da Doença de Alzheimer (DA). Recentemente nosso grupo de pesquisa desenvolveu uma nova metodologia para a obtenção de análogos quirais da Tacrina a partir de cetonas terpênicas, utilizando ácidos de Lewis como catalisadores. Há aproximadamente dez anos os análogos dímeros da Tacrina, conhecidos como bis-tacrina, mostraram maior eficiência para o tratamento da DA comparativamente ao fármaco Tacrina. Desde então, vários compostos dímeros e híbridos contendo o núcleo tacrina, tem sido sintetizados e submetidos a Estudos de Relação de Estrutura e Atividade Biológica. Neste trabalho, realizou-se a síntese de novos híbridos tacrina-lofina, compostos contendo o núcleo tacrina ligados por espaçadores alquílicos com o núcleo lofina (2,4,5-trifenilimidazol). A síntese de interesse envolveu uma reação tetracomponente entre a benzila, um aldeído aromático, acetato de amônio e uma amina precursora que apresenta em sua estrutura o núcleo tacrina ligado a um espaçador alquílico. Também foram sintetizados dímeros da lofina, bis-lofina. Os produtos obtidos neste trabalho foram purificados por cromatografia em coluna e caracterizados por espectroscopia de Ressonância Magnética Nuclear (RMN) de 1H e 13C e medidas de ponto de fusão. Os produtos estão sendo disponibilizados para ensaios biológicos de inibição da enzima acetilcolinesterase (AChE) no Departamento de Bioquímica - Instituto de Ciências Básicas da Saúde –UFRGS. ...
