A depressão maior é o mais comum dos distúrbios de humor, e inclui sintomas comportamentais, psicológicos e fisiológicos. A depressão maior é o mais comum dos distúrbios de humor, e inclui sintomas comportamentais, psicológicos e fisiológicos. Os sintomas emocionais da depressão incluem anedonia, apatia, pessimismo, sentimento de culpa e perda de motivação; os sintomas vegetativos incluem distúrbios do sono e do apetite, perda de libido e retardo do pensamento e da ação. A teoria clássica sobre as bases bioquímicas da depressão considera um déficit funcional dos neurotransmissores monoaminérgicos, como serotonina (5-HT) e noradrenalina (NOR), em certos locais do cérebro. Porém, uma vez que existem tanto problemas de eficácia quanto de segurança e efetividade com os antidepressivos moduladores do sistema monoaminérgico, tem se buscado entender o papel de outros sistemas na doença, dentre eles o glutamatérgico e o imune. A associação entre a ativação do sistema imune e a patofisiologia da depressão é sustentada por estudos clínicos, em ao menos três conjuntos de evidências: níveis elevados de citocinas pró-inflamatórias em pacientes depressivos, os benefícios de imunomoduladorese a indução de quadros depressivos por imunoterapia com IFN-. Nesse estudo, camundongos Balb/c foram submetidos a 7 administrações diárias de IFN-nas doses de 3MIU/kg e 4MIU/kg e seus pesos monitorados durante todo o experimento. Durante os últimos 3 dias de administração de interferon os animais foram observados quanto a presença de comportamento doentio, e após a sétima administração foram submetidos aos testes de preferência por sacarose, suspensão pela cauda (TST) e campo aberto. Após o 6o dia de tratamento, a dose de 4MIU/kg (mas não a de 3MIU) de IFN-α induziu aumento significativo de sickness behavior em comparação com o grupo controle. Não houve diferença quanto ao consumo de sacarose, peso corporal, tempo de imobilidade no teste de suspensão pela cauda ou à distância total percorrida e número de cruzamentos no campo aberto. Os sintomas apresentados no sickness behavior assemelham-se aos sintomas neurovegetativos da depressão, que tendem a aparecer previamente aos sintomas cognitivos e afetivos. Estudos com lipopolissacarídeo (LPS) sugerem que há diferentes mecanismos neurais subjacentes ao sickness behavior e a resposta comportamental tipo-depressiva devido à ativação do sistema imune, e esse estudo estende a observação para o IFN. A variabilidade dos resultados nesse modelo deve-se também a diferenças experimentais. Considerando a relevância terapêutica de prevenir ou atenuar a depressão induzida por IFN a busca por modelos experimentais úteis na procura de alternativas terapêuticas é relevante e o aqui apresentado pode ser um deles. ...
