O presente trabalho tem como objetivo avaliar o efeito da obesidade, induzida por uma dieta hipercalórica, sobre a resposta metabólica de ratas Wistar em diferentes idades. Ratas Wistar de 2 e 15 meses, no início do experimento, foram tratadas por 21 semanas, com dieta padrão (2,95 kcal/g) ou dieta hipercalórica (3,44 kcal/g). Após a morte dos animais por decapitação, foi coletado o sangue troncular para análises bioquímicas e dois pequenos fragmentos de fígado foram coletados: um para análise histopatológica e o outro para extração e quantificação de lipídeos. A técnica de coloração utilizada para a histopatologia foi hematoxilina-eosina. As concentrações séricas de glicose, colesterol total, colesterol HDL e triglicerídeos, bem como as concentrações hepáticas de lipídeos foram dosadas por método colorimétrico e a concentração sérica de insulina por ELISA. A análise estatística destas concentrações foi realizada por ANOVA de duas vias, seguida por Tukey-Kramer (p<0,05). Os dados histopatológicos foram analisados por frequência simples. As ratas que consumiram dieta hipercalórica tiveram maior consumo alimentar tanto em gramas, quanto em calorias e também um maior ganho de peso quando comparados às que receberam dieta padrão, independente da idade. A dieta hipercalórica provocou aumento da glicemia apenas nas ratas jovens. Tanto a dieta hipercalórica quanto o envelhecimento provocaram aumento da concentração sérica de triglicerídeos. As ratas velhas apresentaram redução na concentração de HDL quando comparadas com as ratas jovens. Não foram observadas alterações significativas na concentração de insulina e colesterol total. Quanto ao conteúdo hepático de lipídeos, as ratas velhas apresentaram maiores concentrações de colesterol total e triglicerídeos, mas não houve alterações significativas na concentração de HDL entre os grupos. As ratas velhas que receberam dieta hipercalórica apresentaram maior percentual de degeneração hepatocelular microvacuolar. Conclui-se que a dieta hipercalórica foi eficaz para induzir um modelo de obesidade independente da idade. As ratas velhas se apresentam mais resistentes às complicações decorrentes da obesidade no metabolismo glicídico, no entanto, são mais suscetíveis às alterações no perfil lipídico, tanto sérico quanto hepático. Em vista da complexidade do metabolismo e do próprio envelhecimento, é necessário realizar outros estudos para investigar os mecanismos envolvidos nestes resultados e na fisiologia do metabolismo no envelhecimento, buscando esclarecer as adaptações metabólicas dos indivíduos mais velhos ao meio em que se encontram. ...

The aim of this study is to evaluate the effect of obesity induced by a high calorie diet on the metabolic response of Wistar rats at different ages. Two and 15 months-old female Wistar rats were treated for 21 weeks with standard diet (2.95 kcal/g) or hypercaloric diet (3.44 kcal/g). After the death of the animals by decapitation, the truncal blood for biochemical analysis and two small liver fragments were collected: one for histopathologic analysis and the other for extraction and quantification of lipids. The staining technique was used for histopathology hematoxylin-eosin. Serum concentrations of glucose, total cholesterol, HDL cholesterol and triglycerides and liver lipidic concentrations were measured by a colorimetric method and serum insulin by ELISA. The statistical analysis of these concentrations was performed by two-way ANOVA, followed by Tukey-Kramer (p <0.05). Histopathological data were analyzed by simple frequency. The female rats that consumed high calorie diet had higher food consumption both in grams, as in calories and also a greater weight gain compared to that received standard diet, regardless of age. The high calorie diet caused increased blood glucose only in young rats. Both calorie diet and aging caused an increase in serum triglycerides. The old rats showed a reduction in the concentration of HDL when compared with young rats. No significant changes were observed in insulin concentration and total cholesterol. The hepatic lipid content, the old rats had higher total cholesterol and triglyceride concentrations, but there were no significant changes in HDL concentration between groups. Old rats given high calorie diet showed higher percentage of microvacuolar hepatocellular degeneration. We conclude that the high calorie diet was effective to induce an independent obesity model of age. Old rats are presented more resistant to complications of obesity in glucose metabolism; however, they are more susceptible to changes in lipid profile, both as serum liver. In view of the complexity of metabolism and aging itself, it is necessary to conduct further studies to investigate the mechanisms involved in these results and metabolic physiology in aging, seeking to clarify the metabolic adaptations of older individuals to the environment they are in. ...
