A engenharia de tecidos baseia-se na utilização de biomateirias aos quais as células devem estar aderidas para que a regeneração do tecido possa ocorrer. Um dos principais métodos utilizados na avaliação da capacidade de adesão celular sobre as matrizes nanométricas é a coloração realizada com 4’,2-Diamidino-2-Phenylindole (DAPI). Nesse contexto, o objetivo do presente estudo foi avaliar a influência do tempo de incubação das células-tronco mesenquimais (CTM) sobre as matrizes de Poli(L-ácido láctico-co-ácido glicólico) (PLGA) na adesão celular. Amostras de CTMs foram isoladas da polpa de dentes decíduos e utilizadas entre a 5ª e 7ª passagem da cultura celular. Foram produzidas matrizes de nanofibras de PLGA puro e PLGA contendo 0,2 % de Span-80 e 0,1% de albumina através da técnica de electrospinning. A análise da adesão celular foi realizada após os tempos de 1, 2, 3, 6 e 8 e 10 horas de incubação celular em ambos os grupos de matrizes. Tanto para as matrizes de PLGA puro como PLGA com emulsão, a adesão após 1h e 2 horas se mostrou significativamente inferior aos outros tempos para p<0,05. Nas matrizes somente com PLGA, a adesão nos tempos de 3h, 6h, 8h e 10h não mostraram diferença estatística. Nas matrizes de PLGA com emulsão, o melhor tempo de incubação foi o de 6 horas. Com isso, o estudo mostrou que um período de 6 h de incubação, já seria tempo suficiente para garantir uma boa adesão das células às matrizes de nanofibras de PLGA. ...
