O metabolismo anaeróbico da glicose é utilizado para manter a homeostasia do trifosfato de adenosina, em tecidos e em situações distintas. O lactato gerado em tais condições é utilizado como substrato para a gliconeogênese ou é oxidado e convertido à ATP. Esse estudo verificou a lactacidemia materna, fetal e placentária em ratos, logo após parto normal ou cesáreo, comparando os níveis de lactato de animais obtidos em condições de normoxia e de asfixia intrauterina. Procuramos identificar a origem do lactato fetal, compreender sua importância para o neonato, e comparar os benefícios do parto normal em relação ao cesáreo. Ratas Wistar no 22° dia de gestação submetidas à cesariana. Um corno uterino foi isolado e mantido em solução salina 0,9% a 37°C por 15 min (asfixiado) (n=9) enquanto era realizada a histerectomia do outro corno, obtendo-se neonatos controle (n=8). Alguns controles foram estimulados a respirarem e mantidos a 34ºC por 60 min (n=7). Ao término da asfixia, alguns neonatos foram decapitados imediatamente, enquanto outros, estimulados a respirarem e mantidos a 34ºC por 60 min (n=7). Na coleta do sangue, foram utilizados tubos tratados com fluoreto de sódio (100mM) e centrifugados (2500G, 10min). Para determinar os níveis plasmáticos de lactato utilizou-se o método enzimático colorimétrico com Kit Katal e os resultados expressos em mmol/L. Os resultados indicam que a asfixia intra-uterina (15 min) aumentou significativamente (p<0,05) os níveis de lactato. Os animais submetidos à asfixia intra-uterina demonstraram lactacidemia maior após recuperação (normóxia) de 60 min. Os níveis de lactato dos animais nascidos de parto normal não apresentaram diferença significativa em relação aos neonatos controle de parto cesáreo. Porém, apresentaram diminuição significativa (p<0,05) dos níveis de lactato após 60 min de recuperação (normoxia). Os níveis de lactato materno após PN foram (p<0,05) menores que os obtidos após PC. Asfixia intra-uterina aumentou (p<0,05) os níveis de lactato nas placentas em relação ao grupo controle e PN. Não houve diferença entre os níveis de lactato obtidos de placentas após parto normal ou grupo cesáreo controle. Conclui-se que os asfixiados não foram capazes de reverterem à acidose metabólica, sugerindo que não utilizaram o lactato como substrato energético. Neonatos de parto normal apresentaram habilidade em reduzir os níveis de lactato plasmático, fato que os diferenciou dos neonatos controle de parto cesáreo.
