A bactéria diazotrófica Azospirillum amazonense promove o crescimento de plantas de importância econômica e, portanto, pode ser uma alternativa sustentável à produção agrícola. A bactéria diazotrófica Azospirillum amazonense promove o crescimento de plantas de importância econômica e, portanto, pode ser uma alternativa sustentável à produção agrícola. Visto que o solo é um ambiente variável em relação à disponibilidade de nutrientes, as bactérias que o habitam são adaptadas geneticamente para suportar situações de restrição nutricional. O nitrogênio é um elemento essencial para os seres vivos, portanto trabalhos que visem entender os mecanismos de resposta de A. amazonense à privação desse nutriente são relevantes. As proteínas PII são transdutoras de sinal envolvidas no metabolismo do nitrogênio, são altamente conservadas e bem distribuídas entre os procariotos. Nosso grupo de estudo isolou e caracterizou dois genes parálogos de A. amazonense que codificam para proteínas PII: glnK e glnB. A análise do genoma dessa bactéria (recentemente sequenciado e em fase de anotação) revelou um terceiro gene que codifica para uma proteína PII, localizado a jusante do gene amtB (codifica para um transportador de amônia). O presente trabalho visa entender o papel de glnK2 na resposta de A. amazonense à limitação de nitrogênio e quais seriam os elementos responsáveis por sua expressão. Análises de similaridade e de filogenia mostraram que esse gene é mais relacionado com o gene glnK do que com o gene glnB, portanto foi denominado glnK2. Neste trabalho também se verificou, por análises de similaridade, os possíveis alvos de interação com as proteínas PII. O gene glnK2 está localizado na extremidade de um contig, e, através de técnicas de “caminhada cromossômica”, foram isolados fragmentos de DNA de diferentes tamanhos, que poderão contribuir para um melhor entendimento dos elementos cis-atuantes que regem a transcrição desse gene. Este trabalho também objetiva gerar um mutante de A. amazonense para o gene glnK2 e caracterizar seu fenótipo. A técnica de Cross-over PCR foi utilizada para deletar o gene glnK2. O produto gerado será clonado no vetor pk19mobsacB, que posteriormente será transferido para A. amazonense, para que o gene selvagem seja substituído. ...
