Introdução: A Doença da Urina do Xarope do Bordo (MSUD) é um Erro Inato do Metabolismo caracterizado pelo acúmulo em tecidos e fluidos corporais de aminoácidos de cadeia ramificada (BCAA), Leucina (Leu), Isoleucina (Ileu) e Valina (Val) e de seus α-cetoácidos correspondentes devido a deficiência na atividade do complexo multienzimático da desidrogenase dos α-cetoácidos de cadeia ramificada. O diagnóstico é feito pela detecção no plasma, destes metabólitos em altas concentrações, principalmente Leu. O ácido fenilbutírico vem sendo utilizado de forma eficaz como tratamento para outros distúrbios metabólicos. O objetivo deste trabalho foi avaliar o potencial efeito terapêutico de uma nanoparticula de ácido fenilbutírico em ratos submetidos a um modelo crônico de MSUD. Métodos: O modelo experimental crônico de MSUD foi composto por 4 grupos: grupo controle (salina); grupo ácido fenilbutírico (50 mg/kg) intraperitoneal (I.P.); grupo MSUD (pool de BCAA: 15,8μL/g contendo 190 mmol/L Leu, 59 mmol/L Ile e 69 mmol/L Val) subcutânea (S.C.); grupo MSUD+nano (50 mg/kg de ácido fenilbutírico I.P. e pool de BCAA 15,8μL/g S.C). A dosagem dos aminoácidos no plasma foi feita por HPLC SHIMADZU lc-20ad. A nanopartícula de ácido fenilbutírico foi obtida utilizando Ultra-Turrax T10 basic. Resultados: As concentrações de Leu, Ileu e Val nos grupos Nano + MSUD foram estatiscamente menores quando comparado ao grupo MSUD. Conclusão: O tratamento com nanoparticula de ácido fenilbutírico foi eficaz em diminuir Leu, Ileu e Val em plasma de ratos submetidos ao modelo crônico eMSUD, podendo ser um tratamento promissor nesta doença, que atualmente é de restrição proteica. ...

The objective of this study was to evaluate the potential therapeutic effects of phenylbutyric acid nanoparticle in rats subject to a chronic model of MSUD. Methods: The experimental chronic model of MSUD had 4 groups: control group (saline); phenylbutyric acid group (50 mg/kg) intraperitoneal (I.P.); MSUD group (BCAA pool: 15.8μL/g containing 190 mmol/L Leu, 59 mmol/L Ile and 69 mmol/L Val) subcutaneous (S.C.); MSUD + nano group (50 mg/kg of phenylbutyric acid I.P. and pool of BCAA 15.8μL/g S.C). The amino acid dosage in plasma was made by HPLC SHIMADZU lc-20ad. The phenylbutyric acid nanoparticle was obtained using Ultra-turrax T10 basic. Results: The concentrations of Leu, Ileu and Val in the Nano + MSUD group were statistically reduced when compared to the MSUD group. Conclusion: The phenylbutyric acid nanoparticle treatment was effective in lowering Leu, Ileu and Val in plasma of rats subject to a chronic model of MSUD, and could be a promising therapy for MSUD patients since current treatment is a protein restricted diet. ...
