dc.description.abstractA engenharia de tecidos é um campo multidisciplinar que tem por objetivo o desenvolvimento de substitutos biológicos que possam ser empregados tanto na reparação, quanto na substituição de tecidos ou órgãos danificados. Entretanto, o sucesso no desenvolvimento desses substitutos depende diretamente da escolha do biomaterial a ser empregado para esse propósito. Os poliésteres alifáticos, tais como o poli(ácido lático), poli(ácido glicólico) e seus copolímeros, são amplamente utilizados em pesquisas para aplicações em engenharia de tecidos. Entretanto, esses polímeros apresentam uma alta hidrofobicidade bem como a falta de grupos funcionais que possam interagir com o meio biológico. Nesse contexto, o presente trabalho investigou a síntese e caracterização de poliésteres contendo grupos funcionais que visam permitir uma melhor interação entre o material sintético e o meio biológico. Primeiramente, um poli(L-ácido lático) de alta massa molecular foi sintetizado através da polimerização em etapas, através de um processo alternativo ao empregado industrialmente. O procedimento envolveu a utilização de peneira molecular para a remoção do excedente de água do meio reacional e o polímero sintetizado apresentou uma Mw = 80.000 g.mol-1 com polidispersidade de 2,5 nas condições otimizadas. Em outro estudo, matrizes de microfibras obtidas por electrospinning e filmes de blendas de poli(D,L-lactídeo) de alta massa molecular (PDLLA) e de poli(L-ácido lático) de baixa massa molecular (PLLA), com diferentes proporções entre os polímeros, foram preparadas e suas propriedades avaliadas. As imagens de microscopia eletrônica de varredura (MEV) mostraram a obtenção de matrizes de microfibras ausentes de imperfeições (sem beads). Os experimentos de ângulo de contato para as amostras mostraram que não houve mudanças na hidrofilicidade dos materiais em consequência da adição do PLLA à blenda. Os experimentos de degradação hidrolítica mostraram que a incorporação do PLLA aumentou a taxa de degradação dos materiais. As amostras de microfibras apresentaram um comportamento de polímeros amorfos, enquanto que os filmes mostraram-se semicristalinos a partir de análises de calorimetria exploratória diferencial (DSC). Os ensaios de tensão versus deformação indicaram um aumento tanto no módulo de Young quanto nas tensões máximas de ruptura dos filmes, enquanto que para as microfibras estes efeitos não foram evidenciados. A síntese de um copolímero contendo funções carboxila pendentes à cadeia principal foi realizada através da polimerização em etapas entre o L-ácido lático e o L-ácido málico em diversas proporções. O poli(L-ácido lático-co-L-ácido málico) (PLLALMA) sintetizado apresentou uma Mw = 6.700 g.mol-1, polidispersidade de 1,4 e uma incorporação de L-ácido málico à cadeia principal de 18 % em mol. As análises térmicas desse polímero mostraram uma natureza amorfa, com uma Tg em 43 °C. Esse polímero foi empregado na preparação de nanofibras e filmes, na forma de blendas com o PDLLA. Os ensaios de ângulo de contato mostraram que as matrizes de nanofibras tornaram-se completamente hidrofílicas quando quantidades superiores a 10 % de PLLALMA foram utilizadas na composição da blenda, mas nenhuma diferença foi observada para os filmes. Os ensaios de adesão celular para as matrizes de nanofibras contendo 30 % de PLLALMA adicionado à blenda mostraram que não houve diferença estatística entre essa matriz e o grupo controle. Os experimentos de viabilidade celular indicaram que não houve diferença estatística nas matrizes de nanofibras contendo o PLLALMA quando comparado ao grupo controle. Por fim, poliésteres foram sintetizados a partir de monômeros derivados do glicerol. Os derivados sintéticos do glicerol contendo as funcionalidades azida e p-toluenosulfonila apresentaram-se termicamente instáveis para a utilização em polimerizações com diácidos ou seus derivados anidridos. A utilização de um derivado contendo um grupo benzila na estrutura do glicerol permitiu a obtenção do poli(3-benzilglicerol-co-succinato) linear com uma massa molecular ponderal média de 61.000 g.mol-1 e uma polidispersidade de 4,1. A análise térmica desse polímero mostrou uma Tg em – 2,3 °C, a qual seria adequada para aplicações em engenharia de tecidos. A tentativa de remoção dos grupos protetores através de reação de hidrogenólise com Pd/C resultou em uma fragmentação do polímero, o qual apresentou uma Mw = 5.600 g.mol-1 e polidispersidade de 1,3. No intuito de melhorar as propriedades desse polímero foram sintetizados terpolímeros contendo L-ácido lático, 3-benzilglicerol e ácido succínico, mas apenas polímeros com baixas massas moleculares foram obtidos.pt_BR
dc.description.abstractTissue engineering is a multidisciplinary field which focuses on the development of biological substitutes to be employed in the restoration and substitution of damaged organs and tissue. However, the successful development of these substitutes is directly dependent on the choice of the biomaterial to be employed. Aliphatic polyesters, such as poly(lactic acid), poly(glycolic acid) and their copolymers have been widely used as biomaterials in tissue engineering but they show a high hydrophobicity and a lack of functional groups to interact with the biological medium. Considering this, the present work has investigated the synthesis and the characterization of polyesters containing functional groups capable of allowing better interaction among the synthetic material and the biological system. Firstly, a poly(L-lactic acid) with high molecular weight was synthesized by step polymerization as an alternative for the industrial process. The procedure involved the use of a molecular sieve to remove the excess of water from the reaction medium and the synthesized polymer showed a Mw = 80,000 g.mol-1 with a polydispersity of 2.5 in the best conditions investigated. In another study, the properties of matrices of microfibers prepared by electrospinning and films from blends of a high molecular weight poly(D,L-lactide) (PDLLA) and a low molecular weight poly(L-lactic acid) (PLLA) in different ratios were evaluated. The images from scanning electron microscope (SEM) showed matrices of microfibers without beads. The contact angle measurements exhibited no changes in the hydrophilicity, due to the addition of PLLA to the blends for both specimens. The hydrolytic degradation experiments showed an increase in the degradation rate of the materials, which was caused by the addition of a low molecular weight PLLA. Differential scanning calorimetry analysis exhibited an amorphous behavior for the microfibers samples and a semi-crystalline behavior for the films. The strain x stress measurements indicated an increase in the Young modulus and in the maximum rupture strength for the films, while no changes were observed for the microfibers matrices. The synthesis of a copolymer containing pendant carboxyl groups attached in the chain was performed through step polymerization from an aqueous solution of L-lactic acid and Lmalic acid in different ratios. The synthesized poly(L-lactic acid-co-L-malic acid) presented a Mw = 6,700 g.mol-1, polydispersity of 1.4 and an incorporation of L-malic acid of 18 % mol. The thermal analysis for this polymer showed an amorphous behavior and a Tg = 43 °C. This copolymer was employed in the preparation of nanofibers and films of blends with PDLLA. The contact angle measurements for the nanofibers exhibited a hydrophilic feature when more than 10 % of PLLALMA was used in the blend, while no differences were observed for the films. The cell adhesion essay for the nanofiber matrix containing 30 % of PLLALMA showed no statistical differences between the matrix and the control group. The cell viability experiment indicated that there were no statistical differences in the cell expansion over the matrix containing the PLLALMA in comparison with the control group. Finally, a copolymer from monomers derived from glycerol was synthesized. The derivative monomers from glycerol containing the functional groups azide and ptoluenesulfonyl were shown to be thermally unstable during the polymerization with diacids or anhydrides. However, the use of a derivative containing a benzyl group in the glycerol’s structure allowed the synthesis of the linear poly(benzylglycerol-co-succinate) with a Mw = 61,000 g.mol-1 and a polydispersity of 4.1. The thermal analysis for this copolymer showed a Tg = - 2.3 °C, which is suitable for applications in tissue engineering. The attempt to remove the protecting groups of the polymer through hydrogenolysis in the presence of Pd/C resulted in polymer fragmentation with a Mw = 5,600 g.mol-1 and polydispersity of 1.3. In order to improve its properties, a terpolymer containing L-lactic acid, benzylglycerol and succinic acid was synthesized but with low molecular weight.en
