dc.description.abstractRelevância Etnofarmacológica: Uncaria tomentosa (unha-de-gato) é uma videira de ampla distribuição nas florestas tropicas das Américas, recentemente incluída na Farmacopéia Americana e consta entre as espécies vegetais mais intensamente estudadas nos últimos anos. A espécie tem sido usada tradicionalmente no tratamento de doenças reumáticas, câncer, assim como para alergias e infecções microbianas. As três frações bioativas reconhecidas incluem: alcalóides oxindólicos (FALC), polifenóis (FPOLIF) e derivados triterpênicos do ácido quinóvico (FAQ). Objetivo: O presente estudo tem como objetivo a investigação da atividade antifúngica de extrato bruto e de frações de U. tomentosa frente a leveduras patogênicas. Materiais e Métodos: Foram utilizados o extrato bruto de cascas, frações purificadas de FAQ e FALC, obtidas por técnicas de fracionamento em fase sólida, e a fração FPOLIF, obtida por precipitação. Os teores de triterpenos e de alcalóides oxindólicos pentacíclicos foram determinados por CLAE-DAD, e o de polifenóis totais por Folin Ciocalteau. O ensaio antifúngico de triagem, a determinação da concentração inibitória mínima (CIM) e a avaliação de sinergismo entre as frações purificadas e o fluconazol foram realizados segundo as recomendações do documento M27-A3 da CLSI (Clinical and Laboratory Standards Institute) de 2008. Resultados: No teste de triagem, tanto o extrato bruto quanto as frações causaram inibição de Candida albicans, C. glabrata, C. krusei e C. parapsilosis. Os valores de CIM indicam uma atividade significativa da fração FPOLIF, na faixa de inibição de 7,81 a 15,62 μg/mL frente a C. glabrata e C. parapsilosis. A fração FAQ também se mostrou ativa, porém, em menor escala. Ambas as frações também demonstraram efeito sinérgico com fluconazol, obtendo um resultado de inibição de 0,98 μg/mL frente a C. krusei, uma cepa intrinsicamente resistente a esse antifúngico azólico. Conclusões: A atividade frente a leveduras patogênicas e o forte indício de atividade sinérgica com fluconazol são inéditos para a espécie e abrem uma nova perspectiva de pesquisa sobre as frações de triterpenos e polifenóis de unha-de-gato.pt_BR
dc.description.abstractEthnopharmacological Relevance: Uncaria tomentosa (cat's claw) is a vine widely distributed in tropical forests of South America, recently included in the United States Pharmacopoeia. Moreover, cat’s claw is one of most intensively studied medicinal herbal in recent years. The species has been traditionally used in the treatment of rheumatic diseases, cancer, as well as microbial infections and allergies. The three bioactive fractions recognized include: oxindole alkaloids (FALC), polyphenols (FPOLIF) and triterpenic acid derivatives quinovic (FAQ). Objective: This study aims to investigate the antifungal activity of crude extract and fractions of U. tomentosa against pathogenic yeasts. Materials and Methods: A crude extract from its bark as well the purified fractions FALC and FAQ, previously obtained by fractioning solid-phase techniques, and FPOLIF fraction, obtained by precipitation, were prepared for that intend specifically. The contents of triterpenes and pentacyclic oxindole alkaloids were determined by HPLC-DAD. The total polyphenols content was estimated through Folin Ciocalteau assay. For testing antifungal screening, the determination of minimum inhibitory concentration (MIC) and the assessment of synergism between fluconazole and purified fractions were performed according to the recommendations of the document M27-A3 CLSI (Clinical and Laboratory Standards Institute), 2008. Results: In the screening test, both the crude extract and purified fractions were able to inhibit Candida albicans, C. glabrata, C. krusei and C. parapsilosis growth. Regarding the FPOLIF fraction, MIC values ranging form 7.81 to 15.62 μg/mL against C. glabrata and C. parapsilosis suggested a profitable antifungal activity for that fraction. FAQ fraction was also active but to a lesser extent. Nonetheless, both fractions showed a perceptible synergistic effect with fluconazole, as noticed by growth inhibition with a 0.98 μg/mL concentration against C. krusei, the last one a recognized azole-resistant strain. Conclusions: Both the antifungal activity against pathogenic yeasts and the strong evidence of synergistic activity with fluconazole are first-time reported to Cat’s Claw bioactive derivative as far as we known. This found discloses new research perspectives on the antifungal activity of polyphenols and triterpene fractions of Cat's Claw. Further studies to enlighten the activity extent and antifungal mechanism are in progress.en
