A mucopolissacaridose tipo I (MPS I) é uma doença de depósito lisossomal causada por deficiência na enzima α-L-iduronidase (IDUA). A mucopolissacaridose tipo I (MPS I) é uma doença de depósito lisossomal causada por deficiência na enzima α-L-iduronidase (IDUA). O acúmulo progressivo de glicosaminoglicanos (GAGs) nos mais variados tecidos resulta em diversas manifestações clínicas, incluindo doenças cardiovasculares. São observados o espessamento de válvula e a dilatação da aorta e do coração. A elastina, que é uma proteína estrutural da matriz extracelular, foi descrita fragmentada em aortas de MPS I, associada ao aumento da expressão de cisteíno catepsinas no tecido. Este trabalho teve como objetivo analisar se as catepsinas estão superexpressas também no tecido cardíaco de modelos MPS I. Os corações de 4 camundongos normais e 4 MPS I foram utilizados. Os ensaios de catepsinas foram desenvolvidos utilizando método fluorimétrico, fazendo-se uso de substratos e inibidores específicos. Os resultados mostraram um aumento de 10x na atividade de catepsinas no grupo MPS I comparado ao grupo normal. Dentre as catepsinas testadas (B, K e S), a catepsina B teve significativamente a maior atividade, a catepsina S teve atividade moderada e a K parece não ter envolvimento, pois não apresentou valores significativos. A atividade de caspase-3 foi analisada para verificar quaisquer processos apoptóticos no tecido, embora não tenham sido observadas diferenças significativas de atividade entre os grupos. Adicionalmente, foi constatado número aumentado de macrófagos CD68+ no tecido cardíaco dos animais MPS I. Estes dados sugerem que as catepsinas, especialmente a catepsina B, podem estar envolvidas na patogênese da miocardiopatia dilatada observada na MPS I. ...
