O oxigênio é um gás pouco solúvel em água, o que torna sua transferência em um cultivo aeróbio um fator crítico a ser considerado. O coeficiente volumétrico de transferência de massa (kLa) é fundamental na avaliação da eficiência da transferência de oxigênio e é, por esse motivo, um dos critérios mais importantes no dimensionamento e escalonamento de um biorreator aeróbio. Esse parâmetro é afetado por diversos fatores, como as condições operacionais e as propriedades do meio de cultivo. No presente trabalho, procurou-se identificar se a biomassa e a concentração desta afetam significativamente os valores de kLa em um cultivo de Bacillus megaterium. Para isso, realizou-se um cultivo batelada com a bactéria B. megaterium em um biorreator do tipo tanque agitado de 5 L durante 12 h. Mantiveram-se constantes as condições de temperatura, taxa de aeração, velocidade de agitação e pH do meio. Os valores de kLa foram determinados periodicamente através do método dinâmico. Através desse método obtiveram-se também os valores de taxa de respiração (OUR – Oxygen Uptake Rate) durante o cultivo. A biomassa foi determinada pelo método gravimétrico, nos mesmos intervalos de obtenção do kLa. Através da análise da curva de crescimento celular, observou-se que o micro-organismo foi inoculado em sua fase exponencial de crescimento, começando a sofrer desaceleração metabólica a partir de 6 h e atingido a fase estacionária em 9 h de cultivo. Não se verificou fase lag, nem morte celular durante o cultivo de 12 h. A taxa específica de respiração (qO2) apresentou comportamento típico, com maior atividade metabólica (maiores valores de qO2) durante a fase exponencial e decrescendo juntamente com a desaceleração do crescimento celular. Identificou-se um comportamento anormal na curva de oxigênio dissolvido e de respiração celular (OUR), em 7 h e 8 h. A utilização do método dinâmico para obtenção do kLa levou o sistema a atingir concentrações de oxigênio muito baixas nos períodos de maior respiração, provavelmente inferiores à concentração crítica para B. megaterium. Esse fato ocasionou uma alteração da rota metabólica das células até o restabelecimento do comportamento normal na concentração de oxigênio dissolvido (pO2). Os valores de qO2 reduziram atipicamente para esta etapa do cultivo, enquanto os de oxigênio dissolvido elevaram-se, indicando uma desaceleração inesperada do metabolismo. Esse resultado demonstrou a limitação da aplicação desse método de determinação do kLa para o cultivo estudado. O kLa não foi significativamente influenciado pela presença de biomassa, conforme confirmado por análise de variância. Esse resultado permite a utilização de valores de kLa que atendam aos interesses do cultivo determina-os previamente para o biorreator sem células. Desse modo, elimina-se a problemática da medição e do controle desse parâmetro durante o cultivo de B. megaterium. A constatação do presente estudo permite ainda a desconsideração do fator biomassa em correlações para predição do kLa nesses cultivos. ...
