dc.description.abstractA contaminação por micoplasma em culturas celulares pode causar consideráveis interferências no metabolismo celular, acarretando em um sério problema para os laboratórios de cultura de células. Com o objetivo de avaliar o efeito da contaminação por micoplasma e do tratamento com o antibiótico removedor de micoplasma (MRA), sobre a medida da atividade de hidrolases lisossômicas, foram cultivadas culturas de fibroblastos de indivíduos normais para realização destas medidas. Para tanto, foram selecionadas enzimas de referência do Laboratório de Erros Inatos do Metabolismo do Serviço de Genética Médica do HCPA sendo elas: ß-galactosidase, ß-glicosidase, arilsulfatase A, ß-glicuronidase e hexosaminidase (Total e A). Mediu-se a atividade em fibroblastos contaminados com micoplasma, antes e após adição do MRA. Os resultados foram comparados com a atividade enzimática em fibroblastos controles (sem contaminação) e fibroblastos controles tratados com MRA. Somente a enzima ß-glicosidade não alterou significativamente sua atividade na presença do micoplasma e nem do MRA. A hexosaminidase Total e a ß-galactosidase sofreram interferência significativa na presença do micoplasma e do MRA. A hexosaminidase A e a arilsulfatase A sofreram interferência significativa somente na presença do MRA. A ß-glicuronidase sofreu alteração de sua atividade somente na presença do micoplasma. Estes resultados mostraram que as enzimas testadas comportaramse de maneira diferente frente à pre sença do MRA e/ou do micoplasma, levandonos a avaliar, em um segundo momento, se estes agentes (micoplasma e antibiótico), poderiam alterar a medida da atividade enzimática em cultura de fibroblastos de pacientes com doenças lisossômicas (DL). Para tanto foi realizada a medida da atividade das enzimas: ß-galactosidase, arilsulfatase B (ASB), hexosaminidase A e a-glicosidase. A atividade destas foram medidas em culturas de fibroblastos contaminadas por micoplasma, antes e após adição do MRA. Os resultados destas atividades foram comparados com as atividades enzimáticas em culturas cultivadas na ausência de contaminação. Somente a ASB alterou significativamente sua atividade tanto na presença do micoplasma quanto do MRA. As demais enzimas não sofreram interferência significativa na presença de ambos. Os resultados obtidos nos permitem concluir que em cultura de fibroblastos, tanto a contaminação por micoplasma quanto o uso de um tipo de antibiótico (MRA), podem ser fator de interferência para medida da atividade de algumas hidrolases lisossômicas. Sendo assim, sugerimos que haja a prevenção da contaminação através da detecção do micoplasma bem como o uso de técnicas assépticas e, para aquelas culturas que não podem ser descartadas, faça-se o uso criterioso de antibióticos capazes de remover o micoplasma, como o analisado neste estudo. Dessa maneira, trabalharíamos com um grau de confiabilidade maior nos resultados laboratoriais.pt_BR
dc.description.abstractContamination of cellular cultures by mycoplasma can cause considerable interference with the cellular metabolism and result in serious problems for cell culture laboratories. To evaluate the effects of mycoplasma contamination and treatment with the mycoplasma removal antibiotic agent (MRA) on the measurement of the activity of lysosomal hydrolyses; fibroblast cultures from normal individuals were cultivated specifically to realize these measurements. To do this, we selected reference enzymes from the Laboratory of Inborn Errors of Metabolism maintained by the Service of Genetic Medicine at the HCPA in Porto Alegre, Brazil as follows: ß-galactosidase, ß-glicosidase, arilsulfatase A, ßglicuronidase and hexosaminidase (Total and A). The activity of the fibroblasts contaminated by mycoplasma was measured before and after the addition of the MRA. The results were compared with the enzymatic activity in controls both of non-contaminated fibroblasts and others treated with MRA. The ß-glycosidase enzyme was the only enzyme that demonstrated no significant activity alterations in the presence of either the mycoplasma or the MRA. All the others showed significant changes - the total hexosaminidase and the ß-galactosidase from the presence of the both the mycoplasma and the MRA ; the hexosaminidase A and the arilsulfatase A from the presence of the MRA only; while the presence of the mycoplasma on its own significantly altered the ß-glicuronidase activity. These results prove that the enzymes we tested behave differently when exposed to MRA and/or the mycoplasma. This raises the question as to whether these agents (mycoplasma and antibiotic) could alter the measurement of enzymatic activity in fibroblast cultures from patients with lysosomatic diseases (DL). To resolve this question, the activities of the ß-galactosidase, arilsulfatase B (ASB), hexosaminidase A and a-glicosidase enzymes were measured in cultures of fibroblasts contaminated with mycoplasma, before and after the addition of MRA. The results obtained were compared with the enzymatic activities of uncontaminated cultures. Significant alteration of activity in the presence of MRA and the mycoplasma was observed only in the ABS. The other enzymes demonstrated no significant alteration in the presence of either. The results obtained permit us to conclude that, in a fibroblast culture, both contamination by mycoplasma and the effects of the use of a type of antibiotic (MRA) can interfere in the results of the measurement of the activity of some lysosomatic hydrolyses. Therefore, we suggest that measures be taken to prevent contamination by the detection of the mycoplasma and the employment of aseptic techniques, and, in those cases where the cultures cannot be disposed of, antibiotics capable of removing the mycoplasma should be utilized as analyzed in this study. In this way, greater confidence in the respective laboratory results can be assured.en
